A post-doctoral position is available to study the modification of the DNA replication program using several genomic loci as model systems. We are asking whether reprogramming DNA replication can facilitate nuclear reprogramming during embryonic development. Our hypothesis is that the DNA replication program must be reset before differentiation can occur. One of our ultimate aims is to improve the efficiency of somatic cell nuclear transfer (SCNT) for the derivation of patient-specific human ES cells for gene therapy.
 
Projects include:
 
Global epigenetic remodeling occurs when pluripotent cells are generated from fibroblasts by ectopic expression of four transcription factors. We are studying the mechanism by which temporal replication changes during this transition.
Reprogramming of DNA replication by modification of chromatin structure.
Changes in nuclear organization and DNA replication during B cell development and their role in immunoglobulin heavy chain (Igh) gene rearrangement.
We study DNA replication programs using different approaches including: 
1. Single DNA molecule analysis for replication initiation sites in several gene loci critical for development. 
2. High-resolution microarrays on the genomic scale. 
3. Chromatin immunoprecipitation (ChIP) in combination with microarrays (ChIP-on-chip) to determine the role of histone modifications in the DNA replication program.

The Department of Cell Biology at the Albert Einstein College of Medicine has initiated a special awards program for outstanding candidates. The awards carry a stipend of ,000/year for three years.
